    利用高效液相色谱串联质谱仪(HPLC-MS/MS)建立了一种能够准确定量测定动物源食品中2种万古霉素类抗生素残留量的方法。动物源样品先用甲酸—乙腈溶液提取,再经乙腈饱和的正己烷除脂,然后通过阳离子固相萃取柱净化,最后在多反应监测(MRM)正离子模式下,使用双去氯万古霉素作为内标进行定量分析。结果表明,在20~200 μg/L的线性范围内,目标物万古霉素和去甲万古霉素线性关系良好,相关系数(R2)均可达到0.997以上。在20,50,200 μg/kg 3个添加水平下平均回收率为88.7%~115.1%,相对标准偏差为3.1%~9.4%。方法检出限(LOD)低至1.0 μg/kg,定量限(LOQ)可达到3.0 μg/kg。该方法快速、灵敏、准确、重复性好,适用于动物源性食品中万古霉素类抗生素残留量的测定。

    Abstract:

    A accurate and quantitative method was developed for the determination of vancomycin antibiotics residues in animal food by high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). The samples were extracted by formic acid acetonitrile solution, degreased by acetonitrile-saturated n-hexane, purified by cation solid-phase extraction column, detected in the multi reaction monitoring (MRM) positive ion mode, and quantified by internal standard method using didechloro vancomycin as internal standard. The results showed that the target of vancomycin and norvancomycin had a good linear relationship in the range of 20~200 μg/L, and the correlation coefficient (R2) could reach more than 0.997. The average recoveries varied from 88.7% to 115.1% at the spiked levels of 20, 50 and 200 μg/kg, and the relative standard deviations were in the range of 3.1%~9.4%. The limit of detection (LOD) for the method was as low as 1.0 μg/kg, and limit of quantitation (LOQ) could reach 3.0 μg/kg. This method is rapid, sensitive, accurate, reproducible and suitable for the determination of vancomycin antibiotics residues in animal food.
