Study on extraction and toxicity of lipopolysaccharide from aeromonas hydrophila

    以嗜水气单胞菌脂多糖(LPS)为研究对象,采用热酚水法提取及酶解法、氯仿/甲醇沉淀法纯化,测算LPS产率及样品中蛋白质、核酸含量,SDS-PAGE电泳银染法分析纯度。并通过巨噬细胞Ana-1模型来评价纯化LPS样品的毒性和生物活性。结果显示,纯化后LPS产率为1.22%,蛋白质含量为1.53%,核酸为0.79%,SDS-PAGE主要条带清晰,位置与LPS标准品相同。经纯化LPS样品处理后Ana-1细胞活力降低了21.20%,Ca2+荧光强度为1.67±0.19,与LPS标准品数据无显著性差异(P>0.05)。说明该试验提取、纯化得到的LPS样品纯度高,且有较高毒性与生物活性。

    Abstract:

    To study the lipopolysaccharide (LPS) of Aeromonas hydrophila, LPS was extracted by hot phenol-water method, purified by enzymatic hydrolysis and chloroform/methanol precipitation. The yield of LPS and the contents of protein, DNA were determined. The purity of LPS was analyzed by SDS-PAGE and silver staining.Ana-1 cell model was established to evaluate the toxicity and biological activity of LPS samples. The results showed that the yield of LPS was 1.22%, and the contents of protein and nucleic acid were 1.53% and 0.79% respectively. The main bands of SDS-PAGE were clear and located at same position as the LPS standard. After LPS treatment, the viability of Ana-1 cells decreased by 21.20%, and the fluorescence intensity of Ca2+ was 1.67±0.19, without significant difference compared with LPS standard. The LPS samples extracted from Aeromonas hydrophila had high purity, toxicity and biological activity. This study provided technical support for the development of subsequent extraction and detection methods.
